 血管はその内腔を一層の内皮細胞が覆い、その外側(基底膜側)に動員された壁細胞と総称される血管平滑筋細胞やペリサイトが裏打ちして安定な構造を形成する。上記のように、血管は、生命維持のための基本的な機能として酸素や栄養分および炎症細胞を運搬するとともに、組織形成そのものにも関与する。このため、血管内皮細胞が寿命を迎えると、血管が縮退して、酸素や栄養分が局所まで到達しないだけでなく、血管構造がもろくなり炎症反応を惹起させることになる。これは皮膚においても同様であり、酸素や栄養分の供給が隅々まで行き渡らないだけでなく、皮膚炎症反応を惹起することになる。例えば、血管が縮退からもたらされる血流の減少によって酸素や栄養分が不足した場合には、線維芽細胞の減少によるしわなどの老化現象が報告されており(非特許文献3)、血管構造がもろくなったマウスでは、炎症反応が亢進してしわの形成が促進される知見が得られている(非特許文献4)。さらに、光老化した皮膚部位では、毛細血管の数が減少することが知られており(非特許文献5)、血管内皮細胞の寿命を延長することは、皮膚の状態を正常化し、老化、ひいてはしわ形成を予防することができると考えられる。
 さらに血管は、組織の幹細胞との関連で、組織形成維持の機能を有するため、その破綻、損失や過増生が様々な疾患の原因となる。たとえば、がん、心筋梗塞、脳梗塞などの主要死因となるいわゆる三大疾患は、加齢および生活習慣病の発生とともに増加することは周知であるが、これら三大疾病のいずれにおいても血管異常が観察される。加齢や生活習慣病による血管内外環境因子の変化が、血管に対してストレスを及ぼし、血管内皮細胞に細胞老化を誘導することが解明されてきており、そしてこのことが様々な疾患の病状の進行に深く関わることが判明してきている。例えば、血管老化による血管の破綻は、腎障害、骨粗鬆症、アルツハイマー病、網膜症、勃起障害、肺機能障害など種々の臓器の機能低下をもたらすことが知られている(非特許文献6)。
 正常な細胞は永久に分裂することは不可能であり、一定の回数の分裂後に、不可逆的に細胞分裂を停止する。この現象が細胞老化であり、この細胞分裂により誘導される細胞の老化は、染色体上のテロメア構造の短縮が原因となる(非特許文献7)。テロメアとは、細胞の遺伝情報の中心を担う染色体の末端に存在し、特徴的な繰り返し配列をもつDNAと、様々なタンパク質からなる構造により構成される。細胞が分裂するとともに、このテロメアは短くなっていき、テロメアが一定の長さより短くなると、DNA損傷と認識され、DNA損傷応答経路が活性化されることが、細胞老化で増殖能力が消失する原因とされる(非特許文献8)。血管内皮細胞において、テロメア延長作用を有するテロメアーゼを過剰発現させることで、細胞老化を抑制することができることから(非特許文献9)、細胞老化においてテロメアが重要な役割を果たすことがわかる。
 上記の通り、テロメア長の短縮は細胞老化を誘導するが、様々なストレスが血管内皮細胞の細胞老化を誘導することが判明してきている。このような細胞老化因子としては以下のような病態と分子が知られている。1)高血圧に関わるアンジオテンシンII(非特許文献10)、2)糖尿病の主病態である高血糖(非特許文献11)および糖付加蛋白(非特許文献12)、3)血管内皮細胞のDNA損傷を誘導し、細胞老化を促進すると考えられる、高脂血症や肥満で増加する酸化LDL(非特許文献13)、4)酸化LDLの形成やDNA損傷を介して細胞老化を誘導すると考えられる、喫煙。ここで、上記の1)~4)に記載の病態および生活習慣は、すべて動脈硬化危険因子となり得るが、このような危険因子は、共通して、酸化ストレスを亢進させ、DNA損傷だけでなく、テロメアの短縮を促進することも報告されている(非特許文献14)。さらに、血管の破綻による炎症によって、インターフェロンγやTNFαなどの炎症性サイトカインが高まると、内皮細胞の老化を促進させることも報告されている(非特許文献15)。
 血管内皮細胞の老化を誘導する原因は、以上に限定されるわけではないが、おおむね、加齢や生活習慣/生活習慣病によってもたらされる血流内、および血管外からのストレスが、血管内皮細胞の老化を誘導して、様々な疾患の基礎となる血管異常を誘発する。
 以上を鑑みて、加齢や生活習慣/生活習慣病などの根幹に関わる血管内皮細胞の老化による血管異常を抑制するには、血管内皮細胞におけるテロメア活性を維持させることが必要であると考えられる。血管内皮細胞におけるテロメア活性を持続させることで、加齢や生活習慣/生活習慣病によりもたらされる、種々のストレスに抵抗して、血管内皮細胞老化が抑制され、諸臓器への酸素・養分の運搬、秩序のある炎症細胞の浸潤による抗炎症作用がすみやかに行われ、また組織維持の根幹をなす組織特有の幹細胞の維持がなされて、臓器の維持がなされる。このように、血管内皮細胞におけるテロメア活性を維持できる物質が、内服や、注射、および塗布によって可能になれば、個体の老化を抑制する抗老化剤となり得る。

 本発明の課題は、新規な、細胞寿命延長剤、特に血管内皮細胞寿命延長剤、及びテロメアーゼ活性化剤の提供にある。
 本発明者は、様々な生薬エキスを作用させて検討した実験から、ニッケイ(Cinnamomum)属植物由来の抽出物にはテロメアーゼを活性化する作用があることを見出し、本発明を完成するに至った。
 従って、本願は下記の発明を包含する:
(1)ニッケイ属植物由来の抽出物からなる、細胞寿命延長剤。
(2)前記抽出物がケイ種植物由来である、(1)の細胞寿命延長剤。
(3)前記抽出物がケイシ又はケイヒ由来である、(2)の細胞寿命延長剤。
(4)前記抽出物が水抽出物である、(1)~(3)のいずれかの細胞寿命延長剤。
(5)前記細胞が血管内皮細胞である、(1)~(4)のいずれかの細胞寿命延長剤。
(6)細胞老化の進行を遅延又は停止させ、又は細胞老化状態を改善して細胞寿命を延長させるための、(1)~(5)のいずれかの細胞寿命延長剤の使用。
(7)ニッケイ属植物由来の抽出物からなる、テロメアーゼ活性化剤。
(8)前記抽出物がケイ種植物由来である、(7)のテロメアーゼ活性化剤。
(9)前記抽出物がケイシ又はケイヒ由来である、(8)のテロメアーゼ活性化剤。
(10)前記抽出物が水抽出物である、(7)~(9)のいずれかのテロメアーゼ活性化剤。
図1は、ケイヒ抽出物添加の有無による、HUVECにおけるテロメアーゼ活性の比較を示す。
 ニッケイ属はクスノキ目(Lauraceae)、クスノキ科(Lauraceae)の植物であり、300以上の種が存在し、例えばケイ(Cinnamomum cassia Blume)、クスノキ(C. camphora)、マルバニッケイ(C. daphnoides)シバニッケイ(C. doederleinii)、ヤブニッケイ(C. japonicum)、オガサワラヤブニッケイ(C. pseudo-pedunculatum)、ニッケイ(C. sieboldii)、シバヤブニッケイ セイロンニッケイ(C. verum)、シナモン(C. zeylanicum)が知られる。本発明における細胞寿命延長剤、特に血管内皮細胞寿命延長剤、及びテロメアーゼ活性化剤として好ましくはケイ(Cinnamomum cassia Blume)、特にケイの若枝であるケイシ(桂枝)又はケイヒ(桂皮)由来の抽出物が使用される。ケイの樹皮であるケイヒに由来する抽出物は例えば育毛剤の有効成分として有用であること(特開平10-265350号公報)、Tie2(tyrosine kinase with Ig and EGF homology domain-2)リン酸化活性を有すること(特開2009-263358)は知られるが、それが細胞寿命延長特性、特に血管内皮細胞寿命延長特性、及びテロメアーゼ活性化特性などを有することは全く知られていない。
 上記抽出物は常法により得ることができ、例えばその起源となる植物を抽出溶媒とともに常温又は加熱して浸漬または加熱還流した後、濾過し、濃縮して得ることができる。抽出溶媒としては、通常抽出に用いられる溶媒であれば任意に用いることができ、例えば、水性溶媒、例えば水、生理食塩水、リン酸緩衝液、ホウ酸緩衝液、あるいは有機溶媒、例えばエタノール、プロピレングリコール、1,3-ブチレングリコール、グリセリン等のアルコール類、含水アルコール類、クロロホルム、ジクロルエタン、四塩化炭素、アセトン、酢酸エチル、ヘキサン等を、それぞれ単独あるいは組み合わせて用いることができる。好ましくは、溶媒として水が使用される。上記溶媒で抽出して得られた抽出物をそのまま、あるいは例えば凍結乾燥などにより濃縮したエキスを使用でき、また必要であれば吸着法、例えばイオン交換樹脂を用いて不純物を除去したものや、ポーラスポリマー(例えばアンバーライトXAD-2)のカラムにて吸着させた後、所望の溶媒で溶出し、さらに濃縮したものも使用することができる。
 また上記抽出物として、例えば、ケイヒ抽出液(日本粉末薬品株式会社)等の市販品を用いてもよい。
 本発明で使用される場合、「細胞寿命延長」とは、特に限定されるものではないが、加齢や生活習慣/生活習慣病によってもたらされる血流内、および血管外からのストレスにより引き起こされる細胞老化の進行を遅延又は停止させ、又は細胞老化状態を改善して、細胞寿命を延長させることを言う。
 細胞は特に限定されないが、好ましくは血管上皮細胞である。
 本発明で使用される場合、「テロメアーゼ活性化」とは、テロメアーゼの活性を向上又は促進させることを言い、当該活性は、既知の方法で調べることができる(例えば、Quantitative Telomerase Detection Kit;Allied Biotech,Incを用いる)。
 本発明の細胞寿命延長剤、特に血管内皮細胞寿命延長剤、及びテロメアーゼ活性化剤は、細胞老化に起因する種々の疾患および老化の予防、改善に有効な医薬品または化粧品として利用できる。当該細胞老化に起因する種々の疾患および老化としては、種々の炎症性疾患、免疫性疾患、成人病など、例えば、種々の感染症、関節リウマチ、痛風、高血圧、糖尿病、動脈硬化症、高脂血症、肥満、臓器機能不全などが挙げられる。また、本発明の細胞寿命延長剤、特に血管内皮細胞寿命延長剤、及びテロメアーゼ活性化剤は、細胞老化に起因する皮膚の疾患および老化の予防、改善に有効な医薬品または化粧品として利用できる。当該細胞老化に起因する皮膚の疾患および老化としては、肌荒れ、しわの形成、斑の着色、土色化、たるみの形成、傷つき易くなる、萎縮などが挙げられる。
 更に、本発明は、上記細胞寿命延長剤を上記疾患や老化の予防、改善のため、当該予防、改善を必要とする細胞を含む部位に適用する治療的又は美容的方法も提供する。
 本発明の細胞寿命延長剤、特に血管内皮細胞寿命延長剤、及びテロメアーゼ活性化剤は、その使用目的に合わせて用量、用法、剤型を適宜決定することが可能である。例えば、本発明の細胞寿命延長剤、特に血管内皮細胞寿命延長剤、及びテロメアーゼ活性化剤の投与形態は、経口、非経口、外用等であってよい。剤型としては、例えば錠剤、粉剤、カプセル剤、顆粒剤、エキス剤、シロップ剤等の経口投与剤、又は注射剤、点滴剤、若しくは坐剤等の非経口投与剤軟膏、クリーム、乳液、ローション、パック、浴用剤等の外用剤を挙げることができる。
 本発明の細胞寿命延長剤、特に血管内皮細胞寿命延長剤、及びテロメアーゼ活性化剤中のテロメアーゼを活性化するニッケイ植物由来抽出物の配合量は、用途に応じて適宜決定できるが、一般には阻害剤全量中、乾燥物として0.0001~20.0質量%、好ましくは0.0001~10.0質量%である。
 また、本発明の細胞寿命延長剤、特に血管内皮細胞寿命延長剤、及びテロメアーゼ活性化剤中には、上記ニッケイ植物由来抽出物以外に、例えば、通常の食品や医薬品に使用される賦形剤、防湿剤、防腐剤、強化剤、増粘剤、乳化剤、酸化防止剤、甘味料、酸味料、調味料、着色料、香料等、化粧品等に通常用いられる美白剤、保湿剤、油性成分、紫外線吸収剤、界面活性剤、増粘剤、アルコール類、粉末成分、色剤、水性成分、水、各種皮膚栄養剤等を必要に応じて適宜配合することができる。
 さらに、本発明の細胞寿命延長剤、特に血管内皮細胞寿命延長剤、及びテロメアーゼ活性化剤を皮膚外用剤として使用する場合、皮膚外用剤に慣用の助剤、例えばエデト酸二ナトリウム、エデト酸三ナトリウム、クエン酸ナトリウム、ポリリン酸ナトリウム、メタリン酸ナトリウム、グルコン酸等の金属封鎖剤、カフェイン、タンニン、ベラパミル、トラネキサム酸およびその誘導体、甘草抽出物、グラブリジン、カリンの果実の熱水抽出物、各種生薬、酢酸トコフェロール、グリチルリチン酸およびその誘導体またはその塩等の薬剤、ビタミンC、アスコルビン酸リン酸マグネシウム、アスコルビン酸グルコシド、アルブチン、コウジ酸等の美白剤、グルコース、フルクトース、マンノース、ショ糖、トレハロース等の糖類、レチノイン酸、レチノール、酢酸レチノール、パルミチン酸レチノール等のビタミンA類なども適宜配合することができる。
 次に実施例によって本発明をさらに詳細に説明する。なお、本発明はこれにより限定されるものではない。配合量は質量%である。
実施例1:調製
ケイシ熱水抽出乾燥残分の調製
 ケイシ(Cinnamomum cassia Blume)400.7gに水2Lを加え、3時間加熱抽出を行い、ろ過した。得られた残渣に水2Lを加え、同様の操作を繰り返し、加熱抽出をさらに2回行った。得られたろ液を凍結乾燥し、39.7gの熱水抽出乾燥残分を得た。
ケイシエキスHP-20カラム処理
 上記で得られた熱水抽出物 10gをダイヤイオンHP-20(三菱化学製)カラムにチャージし含水エタノール系で溶出させ、50%エタノール溶出画分を目的溶出画分として得た。
ケイヒ熱水抽出乾燥残分の調製
 ケイヒ(Cinnamomum cassia Blume)20.12gに水200mlを加え、3時間加熱抽出を行い、ろ過して得られた残渣に水200mlを加え、同様の操作を繰り返し、加熱抽出をさらに2回行なった。得られたろ液を凍結乾燥し、1.35gの熱水抽出物乾燥残分を得た。
実施例2:テロメアーゼ活性評価
1)血管内皮細胞(HUVEC)の培養
 通常培地でHUVEC(Promocell)を培養し、P10の細胞を用いて、2% FBSを添加したEBM(登録商標)-2(Lonza)にケイヒ抽出液V(ロットナンバー:080919CE, 日本粉末薬品株式会社)を最終濃度0.01%で添加した培地と、ケイヒ無添加のコントロールの2種類の培地で、6ウェルプレート内で10日間培養した。
2)細胞の回収とテロメアーゼ活性の解析
 試験では、Quantitative Telomerase Detection Kit(Allied Biotech,Inc)を使用した。
 6ウェルプレート内をPBSで2回洗浄し、200μlの1×Lysis Bufferを加えた。30分間氷上で静置したのち、セルスクレイパーを使って細胞を回収した。12,000g(MX-305,TOMY)で5分間、4℃で遠心分離し、160μlの上清を得た。-80℃で保存し、これらをサンプルとして用いた。
 テロメアーゼ活性を定量するに当たり、Control Template TSRを1×Lysis Bufferで0.1 amol/μl、0.02 amol/μl、0.004 amol/μl、0.0008 amol/μlに希釈したものを用いた。
 25℃のインキュベータ内で20分間テロメアーゼ反応をさせたのち、プロトコルに従いテロメアーゼ活性を検出するReal-time PCRを行った(Roche Applied Science LightCycler 480 Instrument, Roche)。
 その結果を図1に示す。ケイヒ抽出液添加培地で培養したHUVECは、ケイヒ抽出液無添加培地で培養したコントロールと比較して、有意なテロメアーゼ活性を示すことが認められた(P<0.05)。従って、ケイヒ抽出液がテロメアーゼを活性化し、その結果細胞寿命を延長できることが示唆された。

Claims (9)

  1.  ニッケイ(Cinnamomum)属植物由来の抽出物からなる、細胞寿命延長剤。
  2.  前記抽出物がケイ(Cinnamomum cassia Blume)種植物由来である、請求項1記載の細胞寿命延長剤。
  3.  前記抽出物がケイシ(桂枝)又はケイヒ(桂皮)由来である、請求項2記載の細胞寿命延長剤。
  4.  前記抽出物が水抽出物である、請求項1~3のいずれか1項記載の細胞寿命延長剤。
  5.  前記細胞が血管内皮細胞である、請求項1~4のいずれか1項記載の細胞寿命延長剤。
  6.  ニッケイ属植物由来の抽出物からなる、テロメアーゼ活性化剤。
  7.  前記抽出物がケイ種植物由来である、請求項6記載のテロメアーゼ活性化剤。
  8.  前記抽出物がケイシ又はケイヒ由来である、請求項7記載のテロメアーゼ活性化剤。
  9.  前記抽出物が水抽出物である、請求項6~8のいずれか1項記載のテロメアーゼ活性化剤。
